Alzheimer’s disease is considered to be the most common kind of dementia in elderly people. It is characterized by the accumulation of a molecule known as amyloid beta in brain. Dr. Mullan’s Alzheimer research has revealed that a molecule CD40L is elevated in the persons suffering from the disease. The studies have shown that the stimulation of this molecule induces increase in amyloid beta levels in the cellular models of disease. Furthermore, the CD40L simulation of cells which are extremely important for defense of nervous system increases pro-inflammatory molecules called Cytokines. One of the cytokines is GM-CSF (granulocyte macrophage colony stimulating factor) that is involved in the inflammation responses in brain.
Dr. Michael Mullan’s Alzheimer research demonstrates that there is a correlation between Alzheimer’s disease and increases in the pro-inflammatory cytokines. The stimulation of CD40L causes an increase in the levels of both amyloid beta and GM-CSF in the AD cell models. The studies show that when these cells are treated with GM-CSF, a time-dependent significant increase in amyloid beta levels occurs. Blocking the GM-CSF reduces the production of CD40L induced amyloid in dose dependent manner. The Roskamp Institute, headed by Dr. Mullan is one of many sites that have been selected to carry out clinical trial on Alzheimer’s disease using vaccine approach.
