Rats are prominent experimental animals in various fields of biomedical research including cardiovascular diseases, cancer research, and industrial toxicology and pharmacology studies. The laboratory rat (R. norvegicus) is more robust than mice for physiological and behavioral studies, and has historically been the model of choice for these applications. However, due to the of the lack of transgenic rats mice were the eminent research animal over the last decades.
The recent availability of rat embryonic stem cells now allows us to make transgenic rats and our line of knock-out rats, and knock-in rats. The technology is similar to that used for gene-targeting in mice, however the culture of rat embryonic stem cells require special conditions. Also, only few rat embryonic stem cell lines have been tested for successful germ-line transmission as this test is time-consuming and expensive.
